Atila OPENCI™ cfDNA Library Preparation Kit
The Atila OPENCI™ cfDNA Library Preparation Kit is a ligation-based system that incorporates a dual UMI (Unique Molecular Identifier) design. This enables precise tracking of individual DNA molecules originating from the same double-stranded template. During ligation, both strands of each double-stranded DNA fragment are tagged with UMIs on both ends prior to amplification, allowing reads derived from the original molecule to be grouped computationally after sequencing.
This dual UMI strategy enables consensus generation by first collapsing reads derived from the same strand into single-strand consensus sequences, followed by comparison of complementary strands to generate a duplex consensus. By requiring agreement between both strands of the original DNA molecule, sequencing and PCR errors can be effectively suppressed, significantly improving confidence in low-frequency variant detection.
This approach provides accurate molecule counting and enhanced analytical sensitivity for applications such as rare variant detection and minimal residual disease (MRD) analysis.
